How to Choose CASA Sperm Counting Chambers?

Many manufacturers produce counting chambers for popular CASA semen analyzers, including IMV, MiniTube, Magapor and ECI/Eacenic. Each brand follows its own set of matching specifications. Even when slides look almost identical, differences in chamber depth, recommended sample‑loading volume or chamber layout can skew your sperm‑test results.

Lots of buyers only check chamber depth when ordering counting chambers, yet overlook critical loading‑volume and chamber‑layout requirements. This is the number‑one cause of inconsistent CASA readings.

Frequently Asked Questions

Q1: Can I swap counting chambers if they have the same chamber depth?

❌ No. Matching depth alone is not enough. Even for the same 20 μm chamber, OEM systems call for different total sample volumes: 3 μL, 3.5 μL and 4 μL. Just because you see the chamber fill up does not mean your results will be correct.

CASA software uses hard‑coded calculation settings. If loading volume or chamber count does not match the factory setup, sperm‑concentration figures will read falsely high or low — you cannot spot this issue visually.

Q2: When do I pick 20 μm versus 10 μm chambers?

20 μm: Mostly used for bovine, equine and other large‑animal semen work

10 μm: Mainly for porcine and ovine semen testing

Results depend heavily on accurate chamber‑depth values. Any depth error changes the optical viewing layer and messes up both motility and concentration outputs.

Q3: Should I pipette sample into each separate chamber?

❌ Never load chambers one at a time. The listed loading volume is the total sample you pipette into the inlet port. Capillary flow fills every chamber automatically. Do not split your sample across multiple wells. Example: For genuine Magapor slides, pipette the full 3.5 μL sample straight into the inlet in one go.

Q4: What is the difference between 4‑chamber, 5‑chamber and 8‑chamber slides? Can I use them interchangeably?

IMV‑Cassou (new‑version Leja)

MiniTube: 4 chambers (A‑D)

Magapor Kubus: exclusive 5‑chamber design (A‑E) ‑ Legacy Leja: 8‑chamber slides (for older machines still in operation)

ECI/Eacenic: compatible slides available in 4/5/6/8‑chamber formats

Varied chamber counts mean different effective viewing areas and software‑recognition rules. A 4‑chamber slide will not run correctly on a Magapor 5‑chamber unit, and the reverse is also true.

Q5: What points should I check when buying compatible counting chambers?

✅ You must match three parameters to your CASA system, no shortcuts allowed:

  1. Chamber Depth
  2. Number of Chambers
  3. Total Sample Loading Volume stated in your OEM user manual

Do not go by the 20 μm mark alone. All three parameters need to fit your CASA instrument to get trustworthy test data.

Why pick ECI/Eacenic counting chambers

We are the direct manufacturer. Our compatible counting chambers cover 4‑, 5‑, 6‑ and 8‑chamber options, with both 10 μm and 20 μm chamber depths. Our slides meet official specs for IMV‑Cassou, Magapor, Minitube and other mainstream CASA analyzers. You get steady, accurate test performance, alongside cost‑saving pricing versus original OEM consumables.
